Intracellular force-generating mechanisms drive tissue-scale mechanical behaviors. The central schematic provides an overview of the major cellular force-generating mechanisms in developing tissues and serves as the basis for the four surrounding panels, which illustrate how these mechanisms generate distinct tissue-scale mechanical behaviors. It includes actomyosin contractility, actin polymerization, cell–cell adhesion and ion transport through membrane channels. These processes generate and transmit forces within and between cells. (A) ATP-dependent ion transport establishes an osmotic pressure difference (ΔP), driving water influx into the lumen. Black radial arrows indicate the outward mechanical effect of the luminal pressure during lumen expansion. (B) Actin polymerization at the tissue edge generates protrusive forces that drive coordinated, directional collective cell migration. Basement membrane turnover controls cell shape
Basement membranes are scaffolds of proteins, like collagen, that give organs shape. During development, basement membranes must adapt to the growing organ. We infer how fast the basement membrane adapts by measuring collagen turnover. We find that perlecan, another basement membrane protein, regulates collagen turnover to control organ shape.
