A ubiquitin chain-feeding mechanism for BRCA1-A - Nature Communications
The BRCA1-A complex cleaves K63-linked polyubiquitin chains at DNA damage sites to control DNA repair. Here, the authors develop a polyubiquitin activity probe and present cryo-EM structures of BRCA1-A bound to it, revealing how avidity and processivity make BRCA1-A a chain-shortening enzyme.

Our evil ORFeome paper is out! In collaboration with @alex-stark.bsky.social, we screened ~4,000 viral proteins and secreted effectors from bacteria and parasites, covering hundreds of diverse pathogens, for phenotypes in human cells.

TRIM21 induces selective autophagy of viruses and bacteria
Rhinesmith et al. describe an intracellular antimicrobial mechanism called “antibody-directed xenophagy” (ADX). In ADX, the E3 ubiquitin ligase TRIM21 prevents infection by intercepting antibody-coated viruses/bacteria in the cytosol and orchestrating their destruction via selective autophagy. The authors hypothesize that ADX may be co-opted for targeted protein degradation.
