Microglia with distinct origins and non-canonical phenotypes coexist within the border-like niche of the brain’s circumventricular organs
Circumventricular organs are specialized brain regions lacking a blood-brain barrier. Van Hove et al. reveal that these regions harbor unique microglia and border-associated macrophage populations that respond to ageing and metabolism and are partly derived from fetal monocytes entering during a perinatal window, which can fully adopt the microglia identity.

A transient MXD4-dependent proliferative pause enables survival of brain micrometastases
García-Gómez et al. identify a transient state, termed the proliferative pause, that enables the survival of micrometastases during vascular co-option by buffering organ-imposed stress. Targeting vulnerabilities associated with this state suppresses metastatic progression and relapse, revealing opportunities for preventive intervention.

Clonal lineage tracing of innate immune cells in human cancer
Liu et al. develop Mitotrek, a method that uses single-cell multi-omics with mitochondrial DNA clonal tracing to map the clonal origins of innate immune cells in human tumors. They reveal that tumor myeloid populations arise from circulating monocytes with pre-existing epigenetic fate biases, suggesting that tumor immunity is shaped before tumor infiltration.

Macrophage-mediated brain-bone marrow crosstalk promotes chronic stress-induced glioma growth
Yang et al. demonstrate that chronic stress accelerates glioma growth by inducing bone-marrow-derived C5aR1+ stress-associated macrophages (SAMs) in tumor-bearing mice. Stress-induced sympathetic nerve activation induces ADRB2+ monocytes to become CD36+ lipid-laden SAMs. SAMs depletion suppresses stress-related glioma tumor growth, and SAM abundance correlates with self-reported stress levels and portended prognosis in patients.

Capturing somatic mutations in single-cell probe-based assays (ie. 10X Flex) is a hard task, especially if you don't know *exactly* the mutations you want to capture, or if your target sequence is a mutational hotspot with many possible alternative alleles. Here's the way we pulled this off. Enjoy!

🤩 new review on CNS perivascular fibroblasts interactions in injury, disease & neuroimmunity - from Natalia Barbara Pikor group @ethz.ch led by Margaux Océane Verdon - check it out! academic.oup.com/jimmunol/art...
Fibroblasts underpin immune-competent niches in the healthy and diseased CNS. Schematic representation of the topological organization of fibroblasts along the vascular tree and across meningeal and brain parenchymal compartments in the context of (A, B) homeostasis, (C, D) infection, (E) autoimmunity, and (F) sterile tissue injury. The provision of cytokines, chemokines, survival factors, and ECM components by local fibroblasts are indicated in each condition. The legend of depicted cell types and vasculature is provided. (A) In the homeostatic CNS, fibroblastic cells include mural cells, perivascular fibroblasts situated abluminal to mural cells, and meningeal fibroblasts. (B) In the steady-state condition, PDPN+ FRC-like cells are limited to DALTs positioned at the intersect of rostral-rhinal, olfactory, and superior sagittal sinuses. In (C) chronic dural infection and (E) autoimmunity, persistent inflammation is associated with TLS formation. TLS are underpinned by FRC-like cells and FDCs providing T and B cell chemoattractants, survival factors, and canonical markers of FRCs. (D) In infections of the brain parenchyma, similar chemokines and FRC markers are upregulated in the absence of TLS formation. (E) In the context of deep gray matter injury, perivascular fibroblasts of the Virchow-Robin space constitute a major source of scar-forming cells. In addition to profuse ECM deposition, scar-forming fibroblasts secrete myeloid cell survival factors and chemokines to drive early and late phase scarring, respectively.
