What is the biological function of the FXR-BICD2-dynein interaction? Our data suggest that at least one role is to promote rapid formation of stress granules, although we suspect this machinery has broader roles in mRNA trafficking under basal conditions (6/n)

Using in vitro reconstitution, we could show that BICD2 is licensed by the FXRs to stimulate dynein movement. We propose that BICD2 is recruited to pre-existing RNP-motor complexes to activate transport (5/n)

FXR1 and FXR2 bind directly to BICD2 and recruit mRNAs to dynein. BICD2 is not required for the FXR proteins to associate with the motor complex, which we found surprising given its conventional designation as a cargo linker (4/n)

In flies, the RNA-binding protein Egl and its partner, the activating adaptor BicD, play a key role in mRNA transport. But Egl only exists in invertebrates, so how is dynein linked to transcripts in mammals? (2/n)

How are mRNAs linked to molecular motors in mammalian cells? In our new preprint, we show that FXR1/2 are RNA adaptors for the microtubule motor dynein. Our findings have also prompted us to rethink how activating adaptors contribute to motor-based transport (1/n)
