Skin-resident CXCR6+ memory CD4+ T cells and dendritic cells form dermal perivascular hubs that drive relapse in allergic dermatitis
Tissue-resident memory T (TRM) cells are heterogeneous, but how pathogenic subsets drive relapse in allergic dermatitis remains unclear. Asahina et al. identify a pathogenic CXCR6+ CD4+ TRM subset sustained through CXCL16-dependent clustering with CD301b+ cDC2s, revealing the TRM-DC niche as a potential therapeutic target for allergic dermatitis.

Malaria durably disrupts macrophage-to-macrophage crosstalk in the spleen
Blood-stage malaria transiently depletes and remodels splenic macrophages, but how distinct subsets recover remains unclear. A recent report by Mauel et al. shows that a yolk sac-derived CD163high red pulp macrophage subset is selectively and durably lost after malaria, disrupting a growth differentiation factor 15-dependent circuit that supports marginal metallophilic macrophages.

A TMEM119-tankyrase axis restrains microglial phagocytosis to maintain memory and enables pharmacological forgetting
Qi and colleagues show that TMEM119 constitutively restrains microglial phagocytosis by sequestering tankyrase from axin and thereby limiting APC-driven actin assembly. Genetic TMEM119 ablation or interference of TMEM119-tankyrase binding by a brain-penetrant peptide enhances synapse elimination and attenuates established fear memory, opening a new way for pharmacological memory manipulation.
