Very proud of this study! Two fundamental processes in gene expression turn out less tightly connected than thought. And their relationship may help explain some surprising vulnerabilities of cancer cells. A big thank you to Agata and Deepshika for years of hard work, and kudos to all collaborators!

Happy to share my PhD work on the regulatory mechanism of E3 ligase RNF213! RNF213 is not a monomer, but a dimer that switches between inhibited and activated states in an ATP-dependent manner. We tracked the activation mechanism and showed how disease mutations can destabilize the inhibited state.

Currently obsessively reading about filaments, multimers, condensates, and DNA... Fascinating how high-affinity DBD:consensus motif interactions are just the tip of the iceberg of more elusive interactions driven partly by DNA patterns, partly by protein homo/heteromultimerisation propensities. 5/5

In the case of FLI1, the cooperative binding to repetitive DNA is necessary for oncogenicity but is not sufficient: cancer (Ewing sarcoma) happens when FLI1 gets fused to a Gln/Tyr-rich domain of EWS that provides additional interactions - but it is the prot-DNA filaments that nucleate everything.

... that are not enough to make two protein copies stick to each other without DNA, but which help cooperatively bind repetitive DNA. Likely a common phenomenon on DNA repeats. On more complex DNA substrates, hetero-interactions between DBDs of different TFs might prevail.

This gives a 'nucleoprotein filament', but the filamentation work is largely done by the repetitive DNA (without DNA the protein is mono- or dimeric). This is similar to what was recently seen with cryoEM for FOXP3 (Sun Hur's papers). Weak & likely fairly adaptable contacts...

Just read this paper about cooperative binding of the DBD of FLI1 to GGAA-repeat DNA. It includes crystal structures with 2x, 3x, & 4xGGAA + EMSAs & fluorescence polarisation, showing that, on repetitive DNA, each FLI1 copy contacts the next & next+1, giving rise to cooperativity &increased affinity
