For 40 years the 8-nt ‘Kozak Sequence’ was thought to mark sites of translation initiation. In a new study, we revise this model by identifying an ~80-nt sequence—the extended Translation Initiation Sequence (eTIS)— that guides ribosomes to correct start sites. 🧵 www.biorxiv.org/content/10.6...

eEF2K regulates pain through translational control of BDNF
Working in sensory neurons, Smith et al. show that the translation elongation regulator eEF2K is capable of inducing the integrated stress response through ribosome-dependent activation of GCN2. This suppresses global protein synthesis, but it enables the preferential translation of the neuromodulator BDNF. Inhibition of this mechanism or BDNF translation diminishes pain-associated behaviors in vivo.
