And the very important PAO1 did its job. Removing intracellular barriers let us recover more - and more diverse - phages. We also discovered that removing intracellular barriers gives us broader access to phage diversity, while surface modifications change which phages we isolate.

So I spent a lot of time removing defence systems and resident prophages from Pseudomonas aeruginosa PAO1 to make it increasingly permissive to phage infection. And eventually we got viPAO1. Officially, “vi” = virus isolation. Unofficially, it has always meant very important👑

This project began with a recurring problem: there are loads of interesting phages out there, but persuading them to actually reveal themselves on the hosts we use in the lab can be… difficult. Eventually we decided the phages weren't the ones who needed to change. The host did.
