Mitochondrial dysfunction in myeloid cells: a central deficit in autoimmune diseases
Autoimmune diseases arise from genetic and environmental factors that disrupt immune tolerance. Recent studies highlight the role of myeloid cell immunometabolism, particularly mitochondrial dysfunction, in driving autoimmunity. Mitochondria regulate energy homeostasis and cell fate; their impairment leads to defective immune cell differentiation, abnormal effector activity, and chronic inflammation. We propose that chronic metabolic stress reprograms myeloid cells, fueling a vicious cycle of cell death and immune activation. Over time, this may induce several states of maladaptation in myeloid cells. Viewing autoimmune disease through a metabolic lens offers new insight into disease mechanisms and highlights potential therapeutic opportunities targeting mitochondrial function to restore immune balance.

Mitochondria and lysosomes in T cell immunometabolism
Metabolic reprogramming and signaling are key orchestrators of T cell immunity. Recent studies have illustrated important roles for intracellular organelles, especially mitochondria and lysosomes, in enforcing T cell metabolism and signaling in response to various extracellular cues. As such, mitochondrial and lysosomal function contributes to adaptive immunity by regulating T cell activation, differentiation, and functional adaptation. In this Review, we discuss how the interplay between organelle biology and metabolism instructs T cell-mediated immunity, with a particular focus on mitochondria and lysosomes. We also summarize how mitochondria and lysosomes, or their crosstalk with other organelles, orchestrate downstream signaling processes and functional reprogramming of T cells. We conclude with a discussion of the pathophysiological outcomes associated with dysregulation of these organelles.

Accuracy versus Perception: a Benchmark of Deep Learning Models for Single-Image Super-Resolution in Microscopy
Virtual super-resolution deep learning methods provide a powerful solution to overcome the physical and temporal constraints of microscopy imaging. Yet, assessing and choosing an ideal methodological strategy complicates their use in life sciences and creates a lack of trust in these methods. Here we propose an objective comparison of nine popular single-image super-resolution (SISR) models in a collection of publicly available microscopy datasets, including cell components like microtubules, endoplasmic reticulum, and actin, using confocal microscopy, SEM, SIM, SMLM and STED microscopy modalities for fixed and live-cell microscopy data. The proposed models will be assessed quantitatively with a collection of metrics in microscopy and computer vision, and qualitatively by experts in the field. The proposed models will be made accessible through open, user-friendly, containerised notebooks. This systematic assessment of SISR approaches will provide a more comprehensive understanding of these methods' performance and contribute to standardising SISR methods in microscopy.

Molecular basis for the regulation of membrane proteins through preferential lipid solvation - Nature Chemical Biology
Static protein structures can capture the association of lipids, but it is unclear whether the association is due to lipids acting as long-lived ligands or the solvation of preferred lipids around the protein. A computational-experimental framework has now shown that for the protein CLC-ec1, it is the change in lipid solvation energies that drives dimerization, with preferred lipids around the protein modulating this driving force.
