Open Access: Revisiting Platelet-Activating Factor in Asthma: An Exploratory Metabolomics Analysis of PAF C-18:1 and Type 2 Inflammation. Noeul Kang, Young Jin Pyung, Deog Kyeom Kim, Cheol-Heui Yun, Hyun Woo Lee 👉 doi.org/10.1111/all.70499 #Allergy_journal journalallergy.com
Kang and Pyung et al., investigate type 2 inflammation mediated by various platelet-activating factors (PAF) in asthma and their association with achieving clinical remission. Only PAF C18:1 was found to be inversely associated with T2 inflammatory markers, in contrast to other PAF species detected in the plasma of asthma patients (alkyl-PAF and lyso-PAF). The authors suggest a structure-dependent relationship, whereby the double bond in PAF C18:1 may alter receptor binding or signaling.

Open Access: Initial OX40–OX40L Signaling Promotes a Skin-Resident Memory T-Cell Reservoir in Atopic Dermatitis. First author: Takayoshi Komatsu-Fujii; corresponding authors: Ryota Asahina, Toshiaki Kogame, Kenji Kabashima Read the article: doi.org/10.1111/all.70493 #Allergy_journal
This study demonstrates the role of OX40-OX40 signaling in pathogenic resident memory T cells (TRM) development using an oxazolone (OXA)-induced atopic dermatitis mouse model. Blockade of the OX40 pathway during the initial inflammatory phase suppressed TRM development and attenuated recall dermatitis. Human lesional atopic dermatitis skin showed a transcriptomic profile consistent with Th2/TRM-like features. These findings support further studies targeting the OX40-OX40L axis for the treatment of atopic dermatitis.

The ɑ-hairpinin of Ara h 1 contains a dominant epitope that is both frequently recognized and blocks a majority of the IgE against this domain. Including this region in tests may improve diagnosis of peanut allergy. The trimeric structure of the vicilin region explains the ability of Ara h 1 to induce anaphylaxis with a single antibody.

Open Access: Need to Assess On-Demand Versus Continuous Treatment in Allergic Rhinitis in ARIA-EAACI Guidelines. Corresponding author: Jean Bousquet Read the article here: doi.org/10.1111/all.70498
There is ongoing discussion on whether patients with allergic rhinitis should receive continuous long-term or on-demand treatment. In this Letter, Bousquet et al., point to the low medication adherence observed in MASK-air studies and by physicians with allergic rhinitis. The study recognizes that this data was obtained from selected populations and that there is a need for dedicated guideline recommendations.

Efficacy of Full and Limited Endoscopic Sinus Surgery in Chronic #Rhinosinusitis With Nasal Polyps: A Randomised Controlled Trial. First author: Paula Virkkula 👉 doi.org/10.1111/all.70438 #Allergy_journal Read more articles: journalallergy.com/Rhinitis-Sinusitis-Upper-Airways
Both Full and Limited FESS result in clinically meaningful relief of disease-specific health-related quality of life (HRQoL) and nasal breathing, effectively improving nasal symptoms and sleep disturbance in chronic rhinosinusitis with nasal polyps (#CRSwNP) patients. Clinically meaningful differences in decreases of SNOT-22 scores (22-item Sino-nasal Outcome Test) between the groups were not found at 12- and 24–month follow-up. Differences in improvement of anosmia, and decreases in polyp load were seen between the groups.

Tezepelumab Reverses TSLP-Driven Immunometabolic Rewiring in Human cDC2s Underlying Pathogenic Th2 and Dysfunctional Treg Responses. First author: Leticia Martín-Cruz; corresponding author: Oscar Palomares 🌟 doi.org/10.1111/all.70470
This study aimed to determine whether thymic stromal lymphopoietin (TSLP) induces a pathogenic immunometabolic program in human type 2 conventional dendritic cells (cDC2s) and whether tezepelumab can directly reverse this process. TSLP rewires human cDC2 metabolism by increasing both glycolysis and mitochondrial oxidative phosphorylation. This metabolic shift drives a pro-type 2 phenotype required to generate pathogenic Th2 cells and functionally impaired FOXP3+ Tregs. Tezepelumab effectively restores normal metabolic and functional properties in cDC2s, inhibiting Th2 polarization and rescuing Treg function.

This study aimed to identify a set of serum biomarkers that robustly distinguish prespecified severe from mild atopic dermatitis groups. Serum proteomic profiling distinguishes severe from mild atopic dermatitis, revealing an epithelial-enriched severity footprint linked to lactate dehydrogenase (LDH)-associated tissue injury and Th2/Th22 inflammation. Machine learning identified a 9-protein biomarker signature that discriminated severity groups with high cross-validation accuracy (AUC = 0.989).

In bronchial biopsies, senescence-associated secretory phenotype (SASP) and p53 pathway enrichment scores are elevated in severe asthma versus non-severe asthma and healthy controls. SASP enrichment is validated in the independent Newcastle severe asthma cohort (NOVA). SASP enrichment is also elevated in nasal brushings of participants with nasal polyps, independently of asthma status. Senescence pathway enrichment associates with exacerbations, daily oral corticosteroid (OCS), lower post-bronchodilator (post-BD) forced expiratory volume in 1 s (FEV1%) and macrophage/oxidative phosphorylation (OXPHOS) signatures, but not with Th2/eosinophil signatures, supporting a non-T2 mechanism.

Cobalt chloride (CoCl2) and cobalt nanoparticles (CoNPs) induce a shared, severity-dependent inflammatory response in cobalt-sensitized skin characterized by activation of interferon signaling, inflammatory pathways (TNF/NF-κβ, IL6-STAT3), apoptosis/unfolded protein response (UPR), cell cycle programs, and glycolysis. Despite comparable nominal exposure, the two cobalt forms exhibit distinct tissue distribution: CoCl2 penetrates deeply and accumulates intracellularly across epidermal and dermal compartments, whereas CoNPs are largely retained in the stratum corneum. Direct comparison reveals divergent transcriptional programs, with CoCl2 preferentially inducing interferon and antiviral responses (e.g., IRF7), and CoNPs promoting keratinization and epithelial differentiation (e.g., KRT25). Together, these findings demonstrate that cobalt speciation and penetration depth shape tissue exposure and downstream immune responses in allergic contact dermatitis.

MOME August article highlight -In Press: Elucidating the mechanism underlying incomplete immune recovery in people living with HIV through CD4⁺ and CD8⁺ T cell multi-omics | #MolecularMedicine #HIV #Multiomics #Metabolomics #Proteomics #Bioinformatics link.springer.com/article/10.1...
Figure 1. Flowchart illustrating patient cohort enrolment and analysis. Participants were initially classified as controls or cases based on their basal CD4+ T cell count, specifically whether they had more
or fewer than 200 CD4+ T cells/µL. After 48 weeks of treatment, the cases were reclassified as either immunological responders (IR) or immunological non-responders (INR) based on whether they had
recovered to 250 CD4+ T cells/µL. This system of classification has been validated in several cohort studies12–15. The subsequent procedure involved the isolation of proteins and metabolites from CD4+ and CD8+ T cells in basal blood samples for each omics analysis. For experimental validation, a separate subset of participants was selected for targeted qRT-PCR analysis of miRNAs (5 IR and 5 INR) and mRNAs (6 controls, 5 IR, and 5 INR) in CD4⁺ T cells.

Metabolites are not genes — avoiding the misuse of pathway analysis in metabolomics - Nature Metabolism
Pathway analysis, originally developed for gene expression data, has been adapted for metabolomics. However, owing to the unique characteristics and constraints of metabolites compared with genes, pathway analysis in metabolomics often yields misleading or nonsensical results.

With over 150 references, this review article provides an overview of the effect of both nutritional and toxic trace elements on allergic diseases. It includes a discussion on the role of trace elements in the pathophysiology of allergies (iron, zinc, selenium, copper, fluorides, manganese, iodine, molybdenum, chromium, cobalt, silicon, nickel and boron) and a summary of reported studies. Overall, additional nutritional supplementation of trace elements is not currently supported for the prevention or management of allergic diseases, unless there is a clinical deficiency.

Open Access: Molecular Characterization of Non-Specific Esophagitis Reveals a Distinct Non-Eosinophilic Phenotype With Fibrotic Traits. First author: Marc Pfefferle; corresponding authors: Hans-Uwe Simon, Thomas Greuter 👉 doi.org/10.1111/all.70364 #Allergy_journal
This study comprehensively characterizes non-specific #esophagitis (NSE) and elucidates eosinophil-independent fibrosis mechanisms. NSE appears to be a distinct fibrotic phenotype of the esophagus, without any considerable overlap with gastroesophageal reflux disease (GERD) and other chronic inflammatory disorders such as eosinophilic esophagitis (EoE) or lymphocytic esophagitis. Our data suggest the development of fibrotic traits independently of inflammatory changes, in particular tissue eosinophilia.
