The figure is organized into four modules: tumor-intrinsic metabolic targets, endothelial cell metabolism and angiogenesis, stromal and myeloid targeting, and rational combination strategies. Tumor-intrinsic metabolic interventions include inhibition of lactate production, lactate transport, glycolytic enzymes, glutamine metabolism, fatty-acid oxidation, and lactylation-associated signaling. Endothelial-directed strategies include anti-VEGF/VEGFR therapies, PFKFB3 inhibition, and modulation of endothelial barrier function. Under normal conditions, epigenetic modifiers such as sirtuins maintain heterochromatin and silence retrotransposable elements (RTEs). DNA damage recruits these epigenetic modifiers to double-strand break sites (DSBs) to support repair. Their incomplete return causes heterochromatin loss and RTEactivation, generating cytoplasmic cDNA that triggers the cGAS-STING signaling
pathway and type I interferon responses, ultimately leading to inflammatory effects. Regarding metabolic reprogramming, SIRT6 deficiency impairs fatty acid oxidation and promotes glycolysis in vascular smooth muscle cells, while simultaneously affecting hepatocytes to exacerbate lipid metabolic disorders. Metabolic remodeling further reinforces inflammation: macrophages from patients with coronary artery disease exhibit excessive glucose uptake and enhanced glycolysis, promoting mitochondrial ROS.

(A) The Failed Linear Model: Single-Target Inhibition. Traditional approaches view AD or 
cancer as a linear cascade driven by a single molecule (e.g., Aβ). Interventions (red “×”) bluntly 
inhibit one enzymatic node or clear the toxic product. This fails due to network robustness, 
redundancy, and compensatory feedback, leading to off-target toxicity.
(B) The APP Signaling Network: A Dynamic, Pathological Attractor. The core depicts the 
APP network as a dynamic, circular system that can become trapped in a disease “attractor state.” 
Inputs—Tissue Context, Oncogenic Drivers, Aging Stresses, and the Immune-Metabolic Milieu—
converge on the central APP Signalosome, regulated by lysosomal function and organelle contact 
dynamics. Secretase-mediated release of bioactive fragments feeds back to reinforce the pathological state. In AD, this cycle stabilizes a Neurodegenerative Attractor; in cancer, an Oncogenic Attractor.
(C) The Network-Correcting Therapeutic Paradigm. New multi-pronged strategies are required: (1) Spatiotemporal Precision & Drug Repurposing (e.g., BACE1 inhibitor repurposed for brain metastasis); (2) Multi-Target Network Pharmacology (AI-designed combinations of mild BACE1 modulator, gamma-secretase modulator (GSM), and lysosomal enhancer); (3) Cell-Type-Directed Combination Therapy (e.g., letrozole + irinotecan for cell-specific network correction in AD). Schematic representation of the integrated cell- and mouse-based experimental design used to investigate the role of GNMT in modulating methotrexate (MTX)-induced metabolic remodeling. In cell-based studies, liver-derived cell lines with or without GNMT expression were treated with MTX and subjected to stable isotope tracer analyses to assess mitochondrial one-carbon metabolism, glycine cleavage system (GCS) 
activity, and central carbon metabolic pathways. In parallel, wild-type (Gnmt⁺/⁺) and GNMT knockout (Gnmt⁻/⁻) mice were administered low-dose MTX or vehicle control over a long-term treatment period. Mice 
were subsequently fed defined diets containing stable isotope tracers, including L-[2,3,3-²H₃] serine, or [2¹³C] glycine, to enable in vivo metabolic tracing. Bone marrow, liver, and plasma samples were collected to quantify isotopic enrichments in folate-mediated one-carbon metabolites, deoxynucleotide synthesis, DNA methylation, glycolysis, and TCA cycle intermediates using gas chromatography–mass spectrometry (GC–MS).

The unsupervised Gaussian Mixture Model (GMM) applicable to dataset with homogenous distribution was applied to 2.5-3 million FLIRR data points arising from normal control tissue sections. (B) The GMM clustered the normal FLIRR datapoints into 2 clusters: GMM full 1 and 2. This approach allowed us to determine a threshold for FLIRR score (≥0.4) for (B) normal vs (≤0.4) (D) PCa. Furthermore, 
when we applied the GMM to a suspected tumor (MAD20-00512 X1-3, Fig. 1C), diagnosed by H&E as “normal” showed predominance in distribution of the lower FLIRR values as compared to all the normal tissue sections (Fig. 4F), underscoring the potential of FLIRR over H&E in detecting a missed cancerous lesion. However, further pathological correlation is required.
