How do you get a molecule to tie itself into a knot on purpose? David Bakker in my lab used DNA’s helical structure to drive self-threading. We then pulled it with optical tweezers, where the trefoil ssDNA knot tightens elastically, leaving only ~10 nt in the tight knot. doi.org/10.1002/anie...

Linker histone H1.0 loads onto nucleosomes through multiple pathways that are facilitated by histone chaperones
Akbari et al. investigated the mechanisms of linker histone H1.0 loading onto nucleosomes. H1.0 directly binds nucleosomes and DNA, while DNA-bound H1.0 is mobile but does not load onto nucleosomes. Linker histone chaperones facilitate H1.0 loading through enhancing nucleosome binding, increasing H1.0 mobility, and enabling H1.0 loading through DNA sliding.

Nature Structural & Molecular Biology, Published online: 02 October 2026; doi:10.1038/s41594-026-01895-4Single-molecule imaging reveals diverse, megabase-scale heterochromatin domains in C. elegans.
