Developmentally programmed loss of long-range Polycomb interactions is regulated by cohesin
Distal regulatory elements (DREs), such as enhancers, can regulate genes across megabase-long distances, presumably via coming into close spatial proximity. The establishment of new transcriptional programmes during cell type transitions is associated with widespread rewiring of the spatial organisation of the genome, including gain and loss of chromatin interactions. Extensive effort has been invested into understanding how chromatin interactions are formed during development, yet the mechanisms underlying their developmental loss remain largely unclear. By leveraging chromatin accessibility-assisted footprinting, acute protein degradation and chromatin conformation capture, we show that loss of promoter interactions cannot be explained by reduced binding of sequence-specific transcription factors (TFs). Instead, we identify a subset of interactions that depend on cohesin for programmed developmental disruption. These sites are characterized by high Polycomb enrichment and TF occupancy and engage in strong long-range interactions that undergo extensive differentiation-dependent rewiring. Preventing interaction loss by acute cohesin degradation results in the preferential downregulation of associated genes. Together, these results suggest that cohesin indirectly regulates developmental loss of Polycomb interactions by enabling the acquisition of other potentially regulatory contacts in a process that may shape transcriptional programs during cell type transitions. ### Competing Interest Statement The authors have declared no competing interest. Long before the first grant was frozen, graduate students were noticing a quieter set of contradictions. They were told that teaching matters while watching excellent teachers struggle for recognition. They were encouraged to engage with the public while learning that public scholarship rarely counts toward career advancement. They were told that mentorship is central to academic culture while watching faculty who pour themselves into students get passed over for colleagues with longer publication lists. They entered science hoping to improve society and discovered that the system rewards productivity more reliably than it rewards care.

This part on promotion systems for faculty in academia is especially salient. #AcademicSky
Promotion systems that treat mentorship as service and public scholarship as extracurricular. Evaluation structures that count citations and grant dollars but have no metric for the graduate students you helped retain. Department norms that ask junior scholars, and disproportionately scholars from historically marginalized groups, to carry the relational labor that holds communities together while counting none of it toward their advancement.

University professors generally do not make very much relative to our training levels. Most of us, especially in STEM, could make a lot more in industry (and deal with a lot less bureaucracy). Grant money mostly goes to paying students, postdocs, and staff, and is heavily vetted and scrutinized.

MxA is one of the broadest antiviral proteins mammals make. It’s a big reason most bird flu strains never get a foothold in people. The rare ones that succeed have learned to dodge it. How MxA actually recognizes viral targets at the molecular level is unknown for a bunch of technical reasons.
