3D cell shapes come to the fore! We often focus on the apical cortex but her work reveals how complex 3D cell shapes mean that focus misses many key cell-cell contacts 🧪 www.biorxiv.org/content/10.1...

3D epithelial cell topology tunes signalling range to promote precise patterning
Cell behaviours in multicellular organisms are coordinated via both diffusible molecules and by signals based on direct cell-cell contacts. The mode of cell communication used influences the signalling range. In many developing epithelia, contact-based Notch-Delta lateral inhibition signalling is used to pattern cell fates. While previous work revealed that cells can use protrusions to extend the range of Notch-Delta signalling to alter these patterns, this is not a general feature of epithelia. In addition, it is not known how the complex three-dimensional (3D) shapes of epithelial cells influence cell communication. In exploring this question, we show that epithelial cells at the Drosophila wing margin, which lack basal protrusions, contact different neighbours at different heights along their apico-basal axis, effectively increasing the number of neighbours each cell touches. To quantitatively assess this behaviour, we develop a novel mathematical modelling framework (Multi-layer Signalling Model) to simulate Notch-Delta signalling over data-derived 3D cell topologies. The model predicts that lateral cell surface signalling is essential to tune the spacing between SOPs. In agreement, we show that perturbing cortical stiffness and cell tortuosity in vivo modifies SOP spacing. These results emphasise the importance of 3D cell geometry and topology in fine-tuning signalling range. ### Competing Interest Statement The authors have declared no competing interest. The National Science Board, which oversees NSF, has published recommendations to update the foundation's merit review (which was just updated). Below are the main points. www.nsf.gov/nsb/updates/...
The Board recommends that NSF:

Boost participation and invite expert reviews from a wider range of industry, research institutions, venture capital, and regions of the country.
Emphasize that NSF-funded research must deliver societal benefits, including research that advances the nation's economic competitiveness and national security.
Build NSF's award portfolio at the agency level for greater nimbleness and strategic alignment with national priorities, both current and emerging.
NSF previously announced operational changes to its Merit Review to streamline, clarify, and harmonize agency funding strategies to achieve top scientific and national priorities, similar to those made by the National Institutes of Health. In the coming year, the Board will continue working with NSF leaders to implement NSB recommendations for Merit Review policy changes, such as encouraging verification of previously reported results and pursuing more bold research with high reward potential.

"We can't succeed in a new era with yesterday's playbook," says McCrary. "For our nation to win the race for the future, NSF must develop transformative partnerships with the private sector, states and communities. Together, we can blaze a trail to the next frontiers of science, usher in a Golden Age of American Innovation, and deliver on the promise of science and technology for the benefit of all Americans."

Mechanism of single-strand annealing from native mass spectrometry and cryo-EM structures of RAD52 homolog Mgm101
RAD52, the primary single-strand annealing (SSA) protein in humans, forms undecameric rings that bind ssDNA within a narrow, positively-charged groove. Whether RAD52 anneals two complementary ssDNAs on the same ring in cis, or between two ring- ssDNA complexes in trans, is unknown. Here, we determined cryo-EM structures of Mgm101, a RAD52 homolog from yeast mitochondria, in complexes with ssDNA, a duplex intermediate of annealing, and B-form dsDNA product. In all states, Mgm101 forms a closed nonadecameric ring that binds the backbone of the first ssDNA at the base of the narrow groove. The second complementary strand binds directly on top of the first to form an extended, unwound, and circular duplex intermediate of annealing. A third complex captures apparent B-form DNA product bound to a novel beta-hairpin motif located on top of the Mgm101 ring, above the primary DNA-binding groove. Mass photometry and native mass spectrometry confirm and further elucidate the complexes formed in solution. Altogether, our data reveal the full SSA pathway of Mgm101 and suggest it anneals two complementary ssDNAs on the same ring in cis. Structural conservation with RAD52 suggests it is likely to use a similar cis mechanism of annealing.
