Bacteria synthesize a bacteriocin within the cell membrane | mBio
Bacteria have evolved numerous and diverse pathways to produce and secrete bacteriocins as weapons of bacterial warfare. These molecules are processed inside the cell and then transported across the bacterial membrane for secretion. We recently discovered the TMcins, a distinct class of bacteriocins found across gram-positive bacteria that contain a transmembrane helix and kill target bacteria by forming large pores. Using a Staphylococcus aureus strain that naturally produces TMcin, we show that TMcin production occurs in the cell membrane and reveal how bacteria process a membrane-spanning peptide in a lipid environment. This work expands our understanding of how bacteria produce antimicrobials and provides a model for discovering other membrane-localized natural products.

We characterized enzymes that write and erase acetylation on Asgard histone tails. These enzymes complemented chromatin silencing in eukaryotes showing cross-domain functional conservation. We hypothesize a stepwise model for the emergence of histone-based chromatin regulation. (3/4)
Schematic model depicting the stepwise evolution of histone-based chromatin complexity from an ancestral archaeal state through Asgard archaea to eukaryotes. Histone-based packaging and sporadic hPTMs represent an ancestral archaeal baseline. Asgard lineages increased histone abundance, diversified tail-bearing variants, and rewired pre-existing modifying enzymes, creating a potential proto-epigenetic axis. Eukaryotic elaboration (including readers, complex targeting, four core eukaryotic histones) built upon this Asgard foundation.
