Caspase inhibitors have repeatedly entered clinical trials but have not reached clinical practice due to toxicity and limited efficacy. Groborz and colleagues now show that gasdermin D pores admit otherwise membrane-impermeable inhibitors into pyroptotic cells, turning poor permeability into a mechanism of cell-state selectivity.

I have yet to find an ATP-mimetic compound which 𝘤𝘢𝘯'𝘵 activate GCN2 at certain concentrations, including AMP-PNP, ATPyS, and even ADP. This is all in vitro with purified GCN2.

😰 good Monday morning exercise 😰 My two cents on this - ferroptosis is best understood as a terminal process that begins once lipid oxidative damage exceeds the cell’s buffering and repair capacity. Does it lead to cell death? Clearly yes.
