AlphaFold Database is expanding into pandemic preparedness. A scalable library of ∼13,000 barcoded viral open reading frames from >500 viral species enables functional genomic screens across the virome. Applying this platform identified viral regulators of cellular proliferation, antigen presentation, and interferon signaling. This platform enables systematic exploration of viral protein function.

New preprint on technologies to scale up CRISPR screens. We use them to map 665,856 pairwise genetic perturbations and outline a path to comprehensive interaction mapping in human cells. We also introduce an approach for cloning lentiviral libraries with billions of elements.

Unexpectedly, @jurgjn.bsky.social found that running Alphafold3 predictions for protein interactions can yield ipTM scores that are more predictive of true interactions when run in pools of proteins instead of pairwise predictions. Presumably, this reflects some sort of "competition effect".
