📢 New ME/CFS genetics preprint from the Beentjes/ Khamseh/ Ponting groups NB: Not DecodeME We discover 7 genetic associations to ME/CFS that are independently replicated in UK Biobank and/or All of Us ME/CFS case/control status required multiple lines of evidence www.medrxiv.org/content/10.6...
ME/CFS cases (or controls) were defined using multiple lines of evidence in 2 independent UK Biobank cohorts, and 1 independent All of Us cohort. We needed to restrict genetic ancestry to Europeans to minimise the chance of spurious associations. Next, we used TarGene to estimate the effects of genetic variants on ME/CFS risk. Finally, we replicated 7 variants that were both significant in a Discovery GWAS, and in one of two Replication GWAS.

Extreme fatigue, a racing pulse, shortness of breath, brain fog, aching joints—the symptoms of long COVID can vary widely, change unpredictably, and be difficult to even recognize as signs of the condition. Long COVID also remains difficult to treat, as no standard treatments yet exist.

To develop better care for long COVID, scientists need to understand what drives it. But its complex landscape of symptoms arises from equally complex biological mechanisms that may differ considerably from patient to patient. At Gladstone Institutes, new tools and discoveries are rapidly filling in the blanks.

1) We wrote a new article on cell and tissue analysis using genetic data. For ME/CFS, the results strongly point to neurons. One of the top hits is the medium spiny neuron, located in a region deep inside the brain called the striatum.

Starting September, international students will be limited to four years of study. With most PhD programs lasting 5 or 6 years, this is a huge blow - to the International students themselves and to our ability to recruit the best students we can. This is not how the U.S. remains competitive.

Today is the 250th anniversary of Boston’s mass inoculation against smallpox on July 3, 1776. The city was effectively closed to commerce from July through September 1776. The mass immunization effort was cheered as evidence of true independence. Here's how I described it in my book:
While John Adams and fifty-five men in Philadelphia debated a resolution on independence and the final wording of the Declaration of Independence, the people of Boston declared their own independence from smallpox. The measures taken in both cities provided hope that America would not only be a republic free of a king but also a country free from disease. On July 3, 1776, a day after independence was declared in Philadelphia, the Massachusetts General Court voted to allow inoculation. Despite the dangers of infection, the people of Boston and the surrounding areas cheered the news. Ezekiel Price, a court official and insurance broker, declared “Liberty is given for to inoculate for the small-pox; many begin upon it this afternoon.” Hannah Winthrop described the scene: “Boston has given up its Fears of an invasion & is busily employd in Communicating the Infection. Straw Beds & Cribs are daily Carted into the Town.” She wrote that “Men Women & children eagerly crowding to inoculate is I think as modish as running away from the Troops of a barbarous George was the last year.” Dr. James Warren wrote to John Adams that the “rage for inoculation” had “whirled me into its vortex.…”  Warren and his wife Mercy Otis joined Abigail Adams and her four children who had been invited to be inoculated at Abigail’s uncle’s house in Boston along with Abigail’s sisters Elizabeth Cranch and Mary Shaw and their families. Even though Boston was teeming with the sick, the prospect that the city could conquer smallpox made it "a scene delightful indeed." Cotton Tufts was satisfied that he could now "take my leave of Earth, satisfied that the Day of American Redemption was at Hand."

In April @ukbiobank.ac.uk stopped all researchers from accessing their Research Analysis Platform. In May they indicated they would “provide a more specific timetable in early June”. Until access is restored @decodemestudy.bsky.social analysis is stalled. We will keep all participants informed.
