—Long-Term Outcomes of COVID-19 Multisystem Inflammatory Syndrome in Children (MIS-C): A 4.5-Year Observational Study — MIS-C survivors face markedly elevated long-term multisystem risks, challenging the notion of full recovery #LongCovidKids
RESEARCH IN CONTEXT

Added value of this study
This study evaluated the long-term outcomes of children with MIS-C with follow-up extending to 4•5 years post MIS-C, providing the longest and most comprehensive assessment to date in a large, ethnically diverse urban population. By applying rigorous 1:2 propensity score matching to controls of COVID-19 patients without MIS-C, we generated robust estimates of long-term multisystem risk.
We found that MIS-C is associated with marked increases in cardiovascular, neurological, respiratory, gastrointestinal, and hypertensive disorders, with risk elevations persisting across 4•5 years of follow-up. The markedly higher risks of cardiovascular disorders, hypertension and gastrointestinal disease challenge the earlier view of MIS-C as a transient inflammatory illness with complete recovery. These findings redefine expectations regarding the long-term health trajectory of children affected by MIS-C.
Implications of all the available evidence
Our results support the need for structured, long-term clinical surveillance, including routine blood pressure monitoring, cardiovascular assessment, mental health evaluation, and renal follow-up in patients with COVID-19 related MIS-C. Children with pre-existing hypertension, diabetes, autoimmune disease, or older age at MIS-C onset appear to be at particularly elevated risk and may benefit from more intensive monitoring. These findings underscore the importance of coordinated, multidisciplinary care for MIS-C survivors to mitigate long-term complications and reduce morbidity.

A cereblon-based glue degrader of NEK7 regulates NLRP3 inflammasome in a context-dependent manner
Sylvain et al. identified NK7-902, a potent NEK7 degrader. While NK7-902 effectively degrades NEK7 in human and murine systems, inhibition of NLRP3 activity varies by context.
