Elevated kinase activity in leucine-rich repeat kinase 2 (LRRK2) is linked to Parkinson’s disease; this study shows that its ROC GTPase acts as a nucleotide-dependent molecular switch that regulates activation and autoinhibition and reveals how some but not all PD mutations disrupt this regulatory switch.
