Got delayed out of Seattle and to kill time ended up at the Exquisite Creatures exhibit. They have a paddlefish skull on display (along with many other beautiful creatures). But can anyone tell me how the hell this bone pattern depositing happens?! WILD!

To add to all the lovely videos of non-AI processes multicilia beating: ctenophore multiciliated cells! They can reach up to 1mm long, causing a rainbow light refraction effect with the correct light positioning! Taken by myself at MBL in 2019 embryology. Realtime frame-rate!

Vertebrate #Hedgehog signaling is transduced by transmembrane protein Smoothened (SMO) on the primary #cilium, but what is its mechanism? @reitergroup.bsky.social &co show that SMO & ciliary GPCRs regulate ciliary PKA activity to activate the Hedgehog pathway @plosbiology.org 🧪 plos.io/4v7vdnp
Model for HH-mediated control of ciliary PKA activity. GPR161 recruits the PKA holoenzyme to the primary cilium. There, locally elevated levels of cAMP promote dissociation of PKA-R from PKA-C subunits. PKA-C phosphorylates GLI, promoting processing into GLI-R and repressing HH target gene transcription in the nucleus. When HH binds to PTCH, PTCH leaves the cilium and SMO enters in an active conformation. GRK2/3 phosphorylates active SMO. Phosphorylated SMO directly binds and inhibits the catalytic site of PKA-C. PKA does not phosphorylate GLI, and GLI-A induces the HH transcriptional program in the nucleus.

While everyone in my lab is a bit heartbroken. It’s been a good moment to come together to celebrate how fucking awesome Zara was. I know she meant so much to so many people at ucsf, in pharmacology, cell biology, and synthetic biology, the leading edge community, and really across science.

Zara’s impact on my life will stay with me forever. She was insanely insightful, wonderfully kind, and sassier than myself. I will miss our weekly coffee walks and making her laugh until she coughed. My heart has broken in a new way this week, but I will carry her forward in my heart and career.
