My final postdoc project is out today in Nature! We show microbes co-migrated with humans out of Africa; three independent methods agree, and timing matches archaeological / genetic evidence (Fig. 4c). I don’t think everyone realizes how bad things are for US science: For the fiscal year that just ended, the Trump regime withheld $1.4 billion from the National Science Foundation’s research budget, which is about 1/5 of its primary research budget.

β-Hydroxybutyrate precursor 1,3-butanediol modulates enteric pathogen susceptibility and Th17 responses via commensal bacteria | mSystems
Diet is a key determinant of gut microbial composition and mucosal immune function, yet the microbial mechanisms linking diet-mediated metabolic changes to immune regulation remain incompletely understood. T helper 17 (Th17) cells play central roles in both protective mucosal immunity and inflammatory pathology, making them a critical target of immunometabolic regulation. In this study, we show that 1,3-butanediol (BD) treatment, which leads to elevated circulating β-hydroxybutyrate (BHB) independently of diet, is associated with suppression of intestinal Th17 responses, remodeling of the gut microbiota, and reduced levels of segmented filamentous bacteria (SFB). We further demonstrate that BHB-associated microbiota changes are linked to increased susceptibility to enteric infection. This work provides a mechanistic framework illustrating how metabolic state can influence host immunity through selective effects on commensal microbes. These findings inform future studies of microbiota-mediated immune regulation.

Importantly, we show that in preclinical models a maternal vaccine strategy can protect newborns during this critical developmental window, providing a roadmap for precision targeting of this pathogen in babies without disrupting the early life microbiome.

We found C. diff toxins remodel the early life gut in ways that persist well into adulthood. Using mice, human infant biopsies, and organoids, we show that even without symptoms, exposure to this toxin-producing pathogen leaves a lasting imprint on the developing epithelial layer and immune system.

We have known for decades that C. diff asymptomatically colonizes babies. Because it does not cause disease, we have assumed that it is completely harmless. In this study, we show that asymptomatic colonization in early life does not mean that this pathogen is inconsequential
