We tested the helicase question more rigorously too. An independent gel-based unwinding assay agreed with the single-molecule experiments: the soluble hexamer binds ssRNA, but we still detect no RNA duplex unwinding under the conditions tested.

The RNA-binding story was also strengthened substantially: new AlphaFold3 RNA/DNA predictions, EMSAs, additional magnetic-tweezers controls and ATPase experiments all support oligomerization-dependent engagement of ssRNA by NS3 - a big thanks to @dulinlab.bsky.social and Herod labs!

Yes, it’s very unusual. If I had to guess, it might be related to journal policies that refer to supplementary movies as “supplementary videos.” Perhaps that terminology then gets applied more broadly during copyediting or final formatting?

BFVD v3 contains 5.8M viral protein structures, 16× more than v2; 75% high quality, filling a major gap in AFDB coverage. It fully covers 72.6% of reference proteomes and spans 72.7% of ICTV species. The spread of PJ.2.1 in Spain 🇪🇸 raises an obvious question: What about the rest of Europe? The answer: We largely don’t know. Of 886 SARS-CoV-2 sequences collected in Europe since July 1 and submitted to GISAID, 671 are from Spain. Much of the rest of Europe has gone dark on genomic surveillance.

Great thread on the PJ.2.1 variant situation, what we do & do not know, & where it stands now. Worth emphasizing that a recent Yunlong Cao preprint found PJ.2.1 has very high ACE2 affinity, meaning that, like BA.2.75 or BA.2.86, it has plenty of space to acquire new Ab-evading spike mutations.
