This is it. Reduced funding necessitates collaboration. What “productive” looks like will change, how much science and individual can accomplish will change. We’ll lose out on a lot of science as society, no doubt. But I am choosing to see those changes in a positive light.

Via the expression of a destabilized eGFP downstream of an 8x repeat ERE, seeER drives eGFP transcription in response to estrogen-bound ER. As we validated in HEK cells 🔬, this gives seeER the ability to track the dynamics of estrogen-driven transcription with ligand- and receptor-specificity. 4/10
plots showing dynamic increases, followed by decreases, in GFP+ cell count in cells that express estrogen receptor alpha (ERa) and were treated with estradiol (E2). Cells that express androgen receptors (AR) or that express ERa but were treated with dihydrotestosterone (DHT) show no changes in GFP+ cells over time.

Nevertheless, it has been difficult to track neural responses to estrogens. Prior tools either do not track neural responses, or report neural response without temporal dynamics. To overcome this, we developed a new reporter, neuro-seeER, to track estrogen response dynamics in neurons in vivo. 3/10

In the 🧠, estrogens signal via nuclear receptors. These estrogen receptors (ERs) target estrogen response elements (ERE) in DNA 🧬 to modify neural gene transcription, altering the activity + function of estrogen responsive-neurons, leading to profound changes in physiology + behavior. 2/10

Our task-switching paper is now out at Current Biology! www.cell.com/current-biol... We find that that our brains reset to a task-neutral state between trials, providing flexibility when the upcoming task is uncertain. RNNs also learn this strategy, but only when trained to switch tasks.

I find myself holding seemingly contradictory views. On one hand, this is an extraordinary moment. We are watching discoveries built over decades produce treatments, and things are happening now that I couldn't imagine when I started. It is hard not to look at that and feel enormous optimism.

Kim, Geiger et al. report that early-life stress increases the chromatin modification H3K4me1 and its enzymatic writer, SETD7, in the ventral tegmental area. They show that this form of epigenetic priming is necessary and sufficient for early-life stress to sensitize transcriptional, physiological, and behavioral responses to subsequent stressors.
