Fine for screening clones, not so good when your experiment depend on purity of plasmid. If you want to try to out-cheap me on labware, I spent my PhD dumpster diving for single-use drosophila vials and scrubbing them out by hand for reuse. May the science gods help you if you run a screen.

Doing the periodic cleaning of used plasmid miniprep columns and realized in honor of Earth Day it may be worth reminding you molecular biologists: Plasmid miniprep columns can be cleaned and reused essentially indefinitely! Just add 1 M HCl, soak overnight and wash 3x w/ water. Save $ and plastic

I think it would be a bit of a stretch to put it that way. Drt3b is a kind of "machine for printing poly(AC)". It can only do that. It's hard to see how something like this could manage to write an arbitrary sequence, or even a more complex one, into DNA. /19

Totally agree! I see increasing SI size as a symptom of problematic shifts in publishing norms in the last few decades. Emphasis on chasing impact factors & novelty/splash means every paper has to be a bigger & bigger story, and there's disincentive to space out the content

Don’t hide the good stuff in the SI In 2008, we published a paper on parameterizing force fields for unfolded proteins using NMR data, developing an early HPS model for intrinsically disordered proteins. The paper showed the idea, validation with synthetic data, and applications to real proteins.
