Hu et al. integrate spatial multi-omics to identify stem-like lymphoid niches (SLNs) organized by CD70+ ILC2s and Tpex cells that predict PD-1/PD-L1 immunotherapy responses in hepatocellular carcinoma (HCC). They reveal that Bacillus cereus-PI-PLC increases SLN abundance and synergizes with PD-L1 blockade, highlighting microbiota-driven enhancement of cancer immunotherapy through SLNs therapeutically in HCC.

Rade et al. present a longitudinal single-cell atlas of BCMA CAR-T therapy in multiple myeloma, revealing that cilta-cel drives potent CD4⁺ cytotoxic responses and superior outcomes. Their findings identify plasmacytoid dendritic cells as unexpected BCMA-expressing targets and suggest therapeutic potential in blastic plasmacytoid dendritic cell neoplasm.

Rheumatoid joints contain macrophages with reparative potential, yet inflammation persists. Huang et al. demonstrate that these homeostatic macrophages lose their protective role by undergoing PANoptotic death. Mechanistically, C1q activates the mitochondrial enzyme SARM1, driving NAD⁺ cleavage into cADPR, which precipitates metabolic collapse and PANoptosis. The resulting macrophage death amplifies autoimmune tissue inflammation through the release of pro-inflammatory mediators.

Li et al. show that SPOP mutations and USP7 overexpression promote TREX1 upregulation, leading to increased cytosolic DNA degradation and suppression of cGAS-STING-mediated immune activation. Targeting USP7 enhances the effectiveness of radiotherapy combined with immune checkpoint blockade, providing a new strategy to overcome resistance and improve therapy outcomes.

Xu et al. identify a commensal Corynebacterium-derived lipid that activates IL-17⁺ γδ T cells via T cell receptor and IL-1 signaling, enhancing barrier defenses. This finding highlights the translational potential of a microbe-informed therapeutic strategy at barrier surfaces.

Lei et al. reveal that radiotherapy (RT) elevates mitochondrial copper and induces cuproptosis by upregulating CTR1 and depleting mitochondrial glutathione. Furthermore, BACH1 downregulation confers radioresistance by upregulating MT1E/X-mediated cuproptosis defense. Correspondingly, copper-loaded copper ionophores restore RT sensitivity by enhancing cuproptosis, highlighting a therapeutic strategy to overcome tumor radioresistance.
