Overall, this work illustrates how natural regulatory polymorphisms can diversify bacterial behavior by rewiring how conserved systems interpret host-relevant signals, thereby tuning virulence programs and potentially the epidemic capacity of tuberculosis bacilli. Editor: @heran.bsky.social

Importantly, these signaling differences are not just biochemical details. The study also links them to differences in virulence in mice, with the PhoPR variant from 𝑀. 𝑏𝑜𝑣𝑖𝑠 AN5 being the only one associated with reduced virulence in this experimental setting.

The results show that these variants do not behave equally. Some respond to acidic pH and Zn, some appear weakly responsive, one seems constitutively active, and one variant from an 𝑀. 𝑏𝑜𝑣𝑖𝑠 strain linked to direct human transmission is poorly induced in vitro yet remains responsive in macrophages.

PhoPR is a major two-component regulatory system controlling key virulence functions in 𝑀. 𝑡𝑢𝑏𝑒𝑟𝑐𝑢𝑙𝑜𝑠𝑖𝑠. In this study, the authors identify Zn and Cd as new inducers of PhoPR, alongside acidic pH, and compare how distinct PhoPR variants behave when expressed in the same HN878 genetic background.
