Using fluorescence, we can monitor when the RNAP binds to form the closed complex, and with MT, we can determine when the promoter opens. Correlative measurement! We found it actually takes almost 20 s to perform this transition, with one kinetically dominating transition!

We used this assay to perform accurate #FRET experiment on duplex DNA standards, and could follow near 4000 fluorescent molecules in a single field of view in real-time. We could also monitor hairpin opening dynamic as a function of T°C using this assay.

We established an objective based #TIRF that can illuminate a 210x210 um field of view with flat illumination. To maximize the number of tether on a single field of view while using a single #sCMOS camera, we used a prism-based illumination to spectrally separate different #dyes (eg Cy3/Cy5)

As MT is a surface based technique (we tether the magnetic beads to the surface), a good fluorescence approach should also be surface-based, as total internal fluorescence reflection microscopy (TIRF). However, previous attempts where limited in throughput (<10), limiting applications.

in short, correlative #single-molecule assays (ie combining #fluorescence and #force modalities) are usually low throughput, eg optical tweezers plus fluo. Magnetic tweezers (MT) are great has they enable #high-throughput while keeping spatiotemporal resolution still high.
