We developed Adaptive Polishing for the consistent preparation of ~150 nm thin FIB-lamellae. This was done by using imaging feedback to adjust parameters for each lamella, or Adaptive Milling, rather than applying a fixed recipe. Preprint: doi.org/10.64898/202...

As a showcase we used easymode to solve the in situ structure of filamentous IMPDH – the rate-limiting enzyme in GTP synthesis – at 4.0 Å resolution, and to map the cellular environment of this striking enzyme assembly.

We validated the networks by solving structures for 10 distinct targets (15 maps total). Some directly via segmentation, others by combining the outputs of multiple tools – constraining picks to mitochondria, the nucleus, or the minus end of microtubules, for example.

To train general networks, the first step was assembling a large, diverse training set. In the end it comprised 4,000+ tilt series from 25+ species and 58 unique data sources, covering all common detector types and everything from purified samples to lifted-out tissue.

Our findings reveal a unique splice variant and a trapped substrate intermediate never observed before, offering new insights into proteolytic catalysis. We also show proteasomes cluster from the post-meiotic spermatid stage, with striking human-mouse differences in proteasome organisation!

We discovered giant membrane-less islands of proteasomes in human sperm nuclei, containing 20S and 20S-PA200 populations! We solved their in situ structure at sub-nanometer resolution (up to 6.8 Å) and in vitro at near-atomic resolution (1.8 Å)!

We discovered that the SUN5-Nesprin3 LINC complex forms a hexagonal lattice at the sperm HTCA, which radically reshapes the nuclear membrane locally! Our findings also provide the structural basis for understanding the severe ASS (Acephalic Sperm Syndrome) male infertility phenotype.
