I had such a fun time teaching at the U Michigan Cryo-ET Course this week! I taught quantitative ultrastructure analysis and we made cool figures like these! My course materials are at baradlab.com/michigan_tutorial for anyone to try. Workflow include mesh refinement and thickness measurement!

A #FIBSEM #cryoET drama in 4 acts. Prelude: Aquilos shuttle rod does not reach cryo stage due to mechanical misalignment.
1 of 2 alignment pins on Aquilos load lock has mysteriously disappeared.4 pairs of eyes, 4 hours and one treasure hunt later, the alignment pin resurfaces in the most obscure spot. The stray alignment pin is superglued into place (why isn’t this the default anyway)Load lock back in working order - sample unloading can proceed. Happy end.

Reintro: I'm a neuroscientist interested in how cells use molecular topography to control signal transduction. I use #CryoET and mouse genetic tools to map the topography of synaptic proteins and determine in-situ structures of scaffolding interactions that control said topography.

"Is X-ray crystallography dead?" some asked when cryoEM got momentum. Depends on what you work on. For nucleosomes it certainly looks like it is dead: all structures with nucleosomes released in 2023 were cryoEM structures! @nucleosomepolice.bsky.social guillawme.github.io/insights-fro...
Bar graph showing the number of PDB entries containing a nucleosome released every year since 1997 (one structure). Crystal structures are red bars, cryoEM structures are green bars. In 2023, all PDB entries with a nucleosome were cryoEM structures.

(The big slow-down caused by the quarantines during the COVID-19 pandemic is also very apparent when comparing the bars for 2019 and 2020...)
