Out of 10 IDRs, 8 colocalized indiscriminately, whereas TDP43 and UBQ2 formed spatially distinct condensates. In TDP43 a segment forming a transient α-helix (see work by @nicolasluxfawzi.bsky.social) determined self-recognition specificity as a truncated mutant co-localized with all other IDRs.

The Csat we measured span about two orders of magnitude, and stickier sequences condensed at lower concentration (R = -0.72). See doi.org/10.1002/pro.... for our latest stickiness scale

micDROP (_M_ultivalent _I_DRs forming _C_onstitutive _DROP_lets) consists of IDRs of interest fused to a fluorescent protein and to a mini-ferritin (12-mer) scaffold. We express them in yeast, quantify the dilute phase in 1000s of cells, which in turn enables inferring Csat for a particular IDR.

It took a while, but here we are. Our micDROP paper is out in @natcomms.nature.com, led by @atargt.bsky.social. She asked whether the sequence of a disordered region can self-recognize specifically. She measured this for ten IDRs in living yeast cells. www.nature.com/articles/s41...

In 2017 we showed that homomers evolve on the edge of supramolecular self-assembly. Here we show how to exploit this vulnerability so that ALL homomers can become bona fide drug-🎯. Delighted that Aim 3 of our @erc.europa.eu finally polymerized! Manuscript: 👉 rdcu.be/e27bT

This month's cover highlights the article Mutation-induced filaments of folded proteins are inert and non-toxic in a cellular system by Emmanuel Levy, Hector Garcia-Seisdedos and colleagues. Yeast cells harboring mutation-induced filaments of folded proteins or “agglomerates”. The filaments appear large and disruptive, but surprisingly, Levin et al. reveal they are largely inert and non-toxic, unlike aggregates of misfolded proteins.
