This interaction triggers translational stress even in heterologous cell expression - suggesting that SLFN1-SLFN15 heterodimers form hyperactive nucleases. This relates to our prior finding that SLFN/SLFN dimerization interface evolves rapidly in mammals. #RapidEvolution #GeneticConflicts

Injecting Slfn15 in viable Om (DDK) embryos is sufficient to induce the syndrome and deleting Slfn15 in incompatible embryos rescues the syndrome. Demonstrating that interaction between maternally inherited Slfn1 and zygotically expressed Slfn15 induces the DDK syndrome. #HybridIncompatibility

We were missing the paternal side... (skipping 🤯 paternal epigenetics)... we observed that another SLFN, Slfn15, was abundantly expressed from the paternal allele in incompatible embryos. And guess what ? it is tied to another transposons insertion, ETn/MusD, found only in incompatible genomes !

Injecting Slfn1 transcript is sufficient to induce the syndrome in compatible embryos. Slfn1 endonuclease activity is required and induces global translational stress in syndromic embryos. #devbio #molecularbio DDK maternal factor found, after 30 years ! 🤯 But wait, there is more...

Turns out that one of these SLFN, Slfn1, is specifically induced in oocytes carrying the DDK Om allele. Assembly of the DDK genome showed a unique MERVL insertion upstream of Slfn1. #transposons Deleting the TE abrogates Slfn1 levels and rescues the syndrome! #genetics 🤩

In mammals, SLFN are best know for their role in tRNA cleavage and transcriptional shut down upon viral infection. But SLFN-related nucleases function across the tree of life: in animals, bacteria, viruses... But what was the link with DDK incompatibilities ? #immunity 🧐

Fast forward decades of mouse genetics, including work from Michel and the late great Charles Babinet #pioneers, the syndrome paternal and maternal contributions were mapped to the same 500kb locus, termed Om (Ovum mutant). Om contains a cluster of innate immune factors: the Schlafen genes.

DDK mice were derived from inbred lab strains in Japan. In the 60's the syndrome was identified: when DDK females are crossed to males from other lab strain (Balb/c, Bl/6...) = embryonic lethality ☠️. But, males from other sub-species or wild derived strains are compatible. #HybridIncompatibility
