to keep the channels fully intact, we have purified sarcoplasmic reticulum (SR) vesicles directly from the native muscle tissue (completely detergent-free). and then we did tomography ofc, look at these beautiful RyR1 arrays sitting on the vesicle surfaces!

Over the last week I have had separate discussions with three people being disillusioned by how bad their colleagues are at their job. Large organizations seem worse. I wonder whether having line managers work together with their people, instead of just leading from above, is a good solution.

Ask both what your country can do for your and what you can do for your country. If you can lead a lab, you can lead a community. Vote blue in November to rescue democracy and consider running for local offices. The best way to change the system is to be the system. - Veteran and scientist.

After my postdoc I will likely go back to teach high school. As someone who thoroughly enjoys teaching at that level, I feel like its becoming more and more necessary. The education system is being dismantled. Good science teachers are hard to come by in rural America. Rebuild the foundation

After 48 h, cells were fixed with paraformaldehyde and glutaraldehyde, followed by staining for dsRNA and HA using specific antibodies and reversible saponin permeabilization. Cells were imaged by confocal microscopy, then immediately embedded and processed for EM imaging. Right: Model. ATL2 membrane tethering is critical for the spatial organization of flavivirus genome replication. ATL2 tethering dimers bring ER membranes into close contact, facilitating the trafficking of newly synthesized viral genomes from existing vROs into vROs forming on juxtaposed membranes (top). This close connection forms a microdomain which brings together the necessary factors for nascent vRO formation and shields the cytosol-exposed viral RNA from innate immune sensors. Because membrane tethering is reversible, this also permits the expansion of viral genome replication away from sites of existing vRO biogenesis. In the absence of ATL2 membrane tethering (bottom), viral genomes extruded from the vRO are exposed to the cytosol and more likely to form new vROs on the same membrane, leading to the concentration of sites of vRO biogenesis and increased recognition by RIG-I-like receptors (RLRs) in the cytosol.

In Britain, where the Industrial Revolution began, the journalist Henry Mayhew calculated that hundreds of Londoners made their living gathering bones, rags and cigar ends from the street, as well as dog excrement for tanneries.
Commissions of inquiry and censuses followed and gathered information on working conditions and education.
Politicians, lawyers and bureaucrats debated whether allowing poor people to vote would lead to disaster.

Social science research arose to study these issues in a more rigorous fashion.
Economists studied markets, political scientists studied politics and the state, sociologists studied changing society, and psychologists studied minds. Though these investigations had flaws and were sometimes biased, together they rebuilt our public understanding of what was necessary and possible.”
