A BrainSaw at the Max Planck Institute for Brain Research, Frankfurt. A Nikon 16x water-immersion objective on a 400 µm travel PIFOC allows for fast z-stacks. Samples are imaged and cut in the clear water bath. A Leica VT1000 vibratome is seen to the right with a mounted blade.How BrainSaw works. Samples are embedded in an agarose block, which is glued to a slide and clamped into a water bath. The bath sits on a three-axis stage under a 2-photon microscope; the stage moves the sample for both tile scanning and slicing. The microscope images the exposed face of the block, the vibratome cuts off the imaged layer, and the cycle repeats until the samples have been fully imaged. Data are stitched automatically and are ready for downstream analysis. we do see some signals with WGA, depending the species or cell type actually in some Ichthyos. But we are planning a more concrete screen of multiple lectins across many species in ExM, to better give tools for the community. Obviously the ones who will gain most are people working on non-models

When utilized in literature review, LLMs consistently 1. fail to mention female authors in female-led literatures, 2. insist that men are more influential or more heavily cited when this is contradicted by objective citation counts, and 3. attribute women’s work to hallucinated male scholars.

Hot take but any reporter that makes the actin network visible will make any cell look awesome under the microscope! These Xenopus 🐸 ectoderm cells are expressing an actin tension sensor, and you can see the retrograde flow very nicely. Actin is better than microtubules 😁 #FluorescenceFriday

How can a single cell learn without a brain? We explore this in my paper with @wallaceucsf.bsky.social! We discovered that single cells may learn using molecules similar to those that animal brains use to learn, like CaMKII. Cells can also propagate memory states to their progeny!
