We composited soil cores and lost 49 AM fungal taxa doing it. Sequencing individual subsamples found them. The ones that vanish are rare, patchy, and phylogenetically clustered. Also drying vs freezing made no difference really. Sample at the scale your question lives at. doi.org/10.1016/j.so...

ABSTRACT

Spreadsheets are widely used software tools for data entry, storage, analysis, and visualization. Focusing on the data entry and storage aspects, this article offers practical recommendations for organizing spreadsheet data to reduce errors and ease later analyses. The basic principles are: be consistent, write dates like YYYY-MM-DD, do not leave any cells empty, put just one thing in a cell, organize the data as a single rectangle (with subjects as rows and variables as columns, and with a single header row), create a data dictionary, do not include calculations in the raw data files, do not use font color or highlighting as data, choose good names for things, make backups, use data validation to avoid data entry errors, and save the data in plain text files.

I am seeking a motivated student(s) to examine the role of anthropomorphic change in nectar evolutionary ecology.  Potential projects include: 1) assessing how anthropogenic change affects nectar evolution, 2) quantifying phenotypic selection on nectar traits and assessing the ecological contexts where it is a target of selection, and 3) determining the ecological and genetic contributions to nectar variation to allow a predictive framework for evolutionary change. Project details will be determined with the student. Likely systems will be fireweed (Chamerion angustifolium) or white clover (Trifolium repens). In year one of the two year project, they collected soil samples from four mock ground plots across Canada to experimentally identify best sampling and preservation practices and potential impact of current NFI and alternative soil sampling protocols on the soil biodiversity information provided by eDNA. Specific activities will include completing the analysis of DNA-metabarcoding data (bacteria, fungi, invertebrates) obtained from soil samples collected during year-1 of the project and disseminating the results. Dissemination efforts will involve writing an executive report summarizing key findings and recommendations for integrating eDNA analysis into the NFI framework, as well as preparing a manuscript to share these results with the scientific community. The candidate will be based at one of the three participating CFS research centres (Québec city (QC), Sault Ste. Marie (ON), or Victoria (BC)). The annual salary will be ~$69,000 CAD + benefits.
