Two recent new publications from our laboratory, led by the amazing Jacob Odell and Maggie Elpers, respectively, shed new insights into the role of nuclear #lamins and nuclear #mechanobiology in two different contexts.
Fluorescence images showing that expression of lamin A in triple-lamin-knockout (TKO) cells results in the displacement of LBR from the nuclear envelope to the endoplasmic reticulum. The figure also includes quantification of the nuclear-to-cytoplasmic ratio of LBR under the different experimental conditions.Schematic representation, representative images, and quantification of experiments on mouse bone marrow derived macrophages showing that polarization of macrophages into pro-inflammatory M1-like cells results in increased nuclear stiffness within 24 hours.

Lamins A and C is required to anchor emerin at the nuclear envelope, and mutations in these proteins cause devastating diseases such as Emery-Dreifuss muscular dystrophy (EDMD). In work now available in #JBiolChem at www.jbc.org/article/S002..., we identify a novel lamin A/C-emerin binding site.

It’s been a pleasure working with @apdaria.bsky.social, Julie Heffler, and the Prosser Lab on this study, supported by the Leducq Foundation and the National Institutes of Health (NIH). This research is one step to get us closer to an effective treatment for cardiac #laminopathies and #LMNA-DCM.

How do LMNA mutations cause dilated cardiomyopathy (DCM) and other #laminopathies? In our latest preprint (doi.org/10.1101/2024...), led by the amazing Noam Zuela-Sopilniak and Julien Morival, we show that cardiomyocyte-specific lamin A/C depletion causes severe DCM, consistent with other studies.

What allows lamins A and C to confer much stronger mechanical stability to the nuclei than lamin B? We show that even though all lamins can protect the nucleus from small cell intrinsic forces, only lamin A/C protect the nucleus from large deformations: www.biorxiv.org/content/10.1... 1/n

Mechanosensitive ion channels at the plasma and nuclear membranes allow Ca2+ entry into the cytoplasm and nucleus, activating downstream signalling pathways. NE stretching opens NPCs, facilitating entry of transcription factors (TF) and transcriptional coregulators (TCR) that alter gene expression. Additionally, NE stretching activates Ca2+ channels in the ER, promoting Ca2+ entry into the nucleus. Stretch-induced activation of cytosolic phospholipase A2 (cPLA2) further drives the arachidonic acid (AA) pathway, leading to gene expression changes.
