Metabolic disease risk in individuals with obesity is variable and influenced by so-far-unknown factors. Reinisch and Ghosh et al. uncovered distinct cell populations and transcriptional programs in adipose tissue that are strongly linked to metabolic disease. The findings suggest that visceral adipose tissue plasticity is key to maintaining metabolic health despite excess adiposity.

Morrison and colleagues show long-chain fatty acid oxidation has context-dependent effects on hematopoietic stem cells (HSCs). It is dispensable for HSC function in young mice. It becomes necessary for normal HSC function and hematopoiesis in older mice. A high-fat diet increases fatty acid oxidation, and this impairs HSC function.

An engineered T cell (center) is designed to deliver therapies to the brain. This T cell displays orange synthetic receptors on its surface, which recognize components of extracellular matrix fibers that are only found in the brain. Within the cell, downstream signaling pathways are programmed to initiate transcription and release a therapeutic payload—shown as light blue protein secretions.

Since techniques like scRNA-seq, bulk RNA-seq, 16S sequencing, etc. are so ubiquitous in immunology, I think it's important that everyone have at least a basic understanding of how these data are analyzed. Also, it's very helpful for developing ideas and generating hypotheses. #immunosky
