A BrainSaw at the Max Planck Institute for Brain Research, Frankfurt. A Nikon 16x water-immersion objective on a 400 µm travel PIFOC allows for fast z-stacks. Samples are imaged and cut in the clear water bath. A Leica VT1000 vibratome is seen to the right with a mounted blade.How BrainSaw works. Samples are embedded in an agarose block, which is glued to a slide and clamped into a water bath. The bath sits on a three-axis stage under a 2-photon microscope; the stage moves the sample for both tile scanning and slicing. The microscope images the exposed face of the block, the vibratome cuts off the imaged layer, and the cycle repeats until the samples have been fully imaged. Data are stitched automatically and are ready for downstream analysis. Our results show that ACh already modulates cortical activity before eye opening. Transient cholinergic decorrelation may enable short-timescale plasticity and contribute to the broader shift toward desynchronized activity that prepares V1 for sensory processing.

Using chemogenetics, we show that these relationships have a causal component: cholinergic input from the basal forebrain contributes to the movement-associated reduction in V1 activity and regulates the temporal structure of network events.

Periods of elevated ACh were associated with reduced neuronal correlations and an altered temporal structure of network events in V1. Notably, ACh was also linked to a movement-associated reduction in V1 activity, the opposite of what is seen in adults.

To address this, we combined in vivo imaging in unanesthetized neonatal mice with ACh and calcium sensors. We found that ACh release in V1 closely follows body movements during wakefulness and active sleep, but is not evoked by visual stimuli alone.

4/ We found that moderate-high alcohol doses (~4-6 drinks) activate layer 5 pyramidal neurons in the PFC for up to 2 hours. What drives this activation? Dopamine. Blocking D1/D5 receptors prevented this effect, also replicated in brain slices using a D1/D5 agonist + alcohol.

3/ To answer this, we used mice and tools to record the activity of individual neurons: calcium imaging in awake mice and patch-clamp electrophysiology in brain slices. We focused on layer 5 pyramidal neurons in the PFC, cells that receive inputs from the VTA and project to many other brain regions.

2/ Dopamine released from VTA projection neurons also reaches the PFC. But how does dopamine affect prefrontal cortex activity right after alcohol intake? This matters because it may help explain enhanced social interaction and aggressive behaviors induced by drinking.

The supply of blood to brain tissue is thought to depend on the overall neural activity in that tissue, and this dependence is thought to differ across brain regions and across brain states. However, studies supporting these views have measured neural activity as a bulk quantity and related it to blood supply following disparate events in different regions. Here we measure fluctuations in neuronal activity and blood volume across the mouse brain, and find that their relationship is consistent across brain states and brain regions but differs in two opposing brainwide neural populations. Functional ultrasound imaging (fUSI) revealed that whisking, a marker of arousal, is associated with brainwide fluctuations in blood volume. Simultaneous fUSI and Neuropixels recordings showed that neurons that increase activity with whisking have distinct haemodynamic response functions compared with those that decrease activity. Their summed contributions predicted blood volume across states.Brainwide Neuropixels recordings revealed that these opposing populations coexist in the entire brain. Their differing contributions to blood volume largely explain the apparent differences in blood volume fluctuations across regions. The mouse brain thus contains two neural populations with opposite relations to brain state and distinct relationships to blood supply, which together account for brainwide fluctuations in blood volume.
