📣 Check out the latest paper from our lab out now in @pnas.org 📣 We show that nociceptive sensitization can occur in the absence of pain stimulation. We use the thermal grill illusion, and show that nonresponders show greater central sensitization (secondary hyperalgesia) than responders.

This graph took me 3 years to make. It's perhaps the largest-ever #metaanalysis of #TMS #BrainStim data. It includes my first real scientific discovery 🧵: When you stimulate the hand with a brief electric pulse, your brain and spinal cord are momentarily less excitable: muscle output decreases 1/n
Muscle excitability (y-axis) plot against the time between stimulation of the hand (PNS) and stimulation of the cortico-spinal pathway (TMS) to the muscle (x-axis). The x-axis is in milliseconds, from 1 to 1000 on a log scale; the y-axis is the log10 ratio of muscle excitability with prior hand stimulation versus without. Each blue dot in the figure is a group mean, obtained from 353 papers. The blue line is the random effect meta-analytic mean; where the blue line goes thicker (between 17-64ms, then 100-320ms) there is a significant meta-analytic effect (i.e., the line is below zero, muscle excitability is decreased). The plot includes over 11000 datapoints. Most datapoints are concentrated in the 16-30ms period, which is studied as 'short-latency afferent inhibition' (SAI).

Seminowicz Cognitive control is believed to arise from task-dependent interactions among networks of brain regions. Although several debilitating neuropsychiatric disorders are characterized by cognitive network dysfunction, the neural circuit mechanisms supporting task-dependent network activity are largely unknown. External and internal task demands elicit opposing responses from key cognitive networks, and claustrum projections target regions associated with both network states. We tested if claustrum supports task-dependent network activity in humans using fMRI during tasks with externally and internally driven demands: working memory (n = 420) and autobiographical memory (n = 35). Claustrum activity increased in both tasks. Claustrum exhibited anatomical connectivity with regions representing all implicated networks, and claustrum effective connectivity suggested an excitatory influence on regions in multiple task-associated networks. Task response and connectivity measures differed between the claustrum and regions prominently implicated in directing network states—the anterior insula and pulvinar. These findings establish a role for the claustrum in supporting task-dependent network states subserving cognitive control.
