Challen and colleagues show that the clonal expansion of Dnmt3a-null HSCs in serial bone marrow transplantation assays is not dependent on the loss of DNA methylation functions of Dnmt3a. They identify non-canonical functions for Dnmt3a in the regulation of telomere length and genome stability that restrict the functional potential of HSCs.

Matatall et al. uncover a mutation-specific association between clonal hematopoiesis (CH) and Alzheimer’s disease (AD) in mice and humans. TET2-mutant CH conferred lower odds of AD in humans and reduced AD in a mouse model via enhanced peripheral immune cell infiltration, whereas DNMT3A-mutant CH had no impact on AD outcomes.
