Submicrometre sampling of living cells by macrophages - Nature
Macrophages can sample antigens from living cells through a trogocytosis-like mechanism that routes ingested material away from degradation, a finding that delineates a previously unknown pathway for antigen presentation to CD8 T cells.

Amy C Fan (she/her) @amycfanphd.bsky.social · 29/04/2026
We think this creates a stable depot of antigens. Macrophages may not only present this material to T cells, but also potentially share with other immune cells, with implications for self-tolerance and tumor immunity. Questions? Drop them below! #immunology #cellbiology #macrophages #trogocytosis

What regulates this alternative trafficking and what happens when we disrupt it? We found that SNX27-retromer complex diverts live-sampled material away from lysosomes AND Deleting SNX27 in macrophages decreased T cell priming after live-sampling.

If the antigens are not getting degraded, does this affect macrophages’ ability to activate T cells? Yes. Surprisingly, macrophages that sample live-cells efficiently cross-present that material to prime CD8 T cell activation and proliferation

Phagocytosis of dead cells and endocytosis of soluble material led to accumulation of this antigen in the degradative lysosome. In contrast, we found live-sampled materials avoids lysosomes and is diverted to an alternative compartment negative for canonical endolysomal markers.

What happens once this sampled material enters macrophage? To answer this question, we developed a 10-parameter flow-based vesicle profiling method to track vesicles. In effect, we can now do flow cytometry with the contents of a cell and analyze this entire vesicle pool all at once.

Amy C Fan (she/her) @amycfanphd.bsky.social · 29/04/2026
Macrophage-target cell interactions spontaneously result in a tiny protrusion from the target cell that separates into a submicron vesicle containing cytoplasm. This is a fast process, taking just minutes.
Microscopy stills showing green target cell material stretching into a red macrophage and subsequently separating into a discrete green vesicle.

Amy C Fan (she/her) @amycfanphd.bsky.social · 29/04/2026
How does this happen? The antigen-containing vesicles were SO small that conventional microscopy could not capture this process. So we turned to Nikon NSPARC and lattice light-sheet microscopy. This revealed…

Instead, we found that: ✅ Sampling is contact-dependent ✅ Sampling is caspase-independent ✅ Target cell remains alive

Amy C Fan (she/her) @amycfanphd.bsky.social · 29/04/2026
When we put macrophages and highly viable cells together, we saw that macrophages could continuously sample cytoplasmic material from living, healthy cells. This is not: ❌ Endocytosis of soluble particles ❌ Traditional phagocytosis of dying cells.

Amy C Fan (she/her) @amycfanphd.bsky.social · 29/04/2026
We know that the immune system can sample dead/dying cells to determine what is “you” vs “not you.” But -- how does the immune system gather information without destroying cells?
