🧬 Preprint! 🧵 Our goal is to genetically manipulate every embryonic germ layer to enable high throughput in vivo studies. We previously showed mouse ectoderm can be targeted by amniotic injection, enabling cell-specific manipulation, lineage tracing, and knockouts. But how to reach mesoderm?
Schematic of method developed and application. At embryonic day 7.5, a lentiviral library carrying unique 30-nucleotide barcodes and a fluorescent reporter is injected into the mouse exocoelomic cavity. An E8 embryo cross-section shows that this cavity is continuous with the intraembryonic coelomic cavity, allowing the virus to reach and barcode mesodermal progenitors, including lateral plate and paraxial mesoderm.

Just published! IStrons are a unique class of transposable elements that pack three distinct functions in one nucleotide sequence. In this work, we used pooled library mutagenesis and high-throughput sequencing to systematically map the sequence and structural features that govern these functions.

Finally - the 🧵! So, the piRNA pathway defends the animal germline from transposons. But most of what we know comes from narrow developmental windows like late oogenesis, where it's easiest to study. We asked if the pathway is the same across development. It is not! /+

Happy to share our new preprint on the discovery of the autonomous form of the D. melanogaster transposon previously known as Hopper. We characterize its distribution and related elements, including one homolog showing remarkably extensive horizontal spread across Drosophila species.

Calling the hive mind: Has anyone encountered an unusually high number of rDNA-associated reads in a 10x v4 scRNA-seq library? We have millions of reads mapping to Drosophila rDNA, multimapping/low MAPQ, with strong enrichment at specific rDNA regions. Curious if others have seen something similar!

I find it a bit concerning that the authors went to journalists after the rejection, even before the preprint was posted. Also, synthetic biology does not strike me as a field of “underdog scientists”; if anything, it seems like exactly the kind of field where results can easily be overhyped.

Gregory J. Hannon passed away in April 2026 at the age of 61. A towering figure in modern molecular biology, Greg influenced remarkably diverse areas of science. His work reshaped our understanding of gene regulation, advanced the discovery of tumor suppressors that regulate the cell-cycle machinery, and uncovered fundamental mechanisms of RNA interference. By establishing core principles of small RNA biology, he not only transformed our mechanistic view of RNA-silencing pathways but also pioneered functional genomics approaches that revolutionized basic and biomedical research in mammalian systems.
