Exciting news: @uwgenome.bsky.social will be hiring for two tenure-track faculty positions this year, one open rank and one assistant professor. Both are “Genome Sciences” interpreted broadly so we welcome applicants working in a variety of fields and methods. Ad will be out shortly.

Early on a program officer told me that my grant was not a contract and the best thing to do was “follow the science”.

So we have connected vertebrate MSTO1 to TRiC (this connection was reported in Drosophila, although there lower TRiC via reduced MSTO1 caused mitotic arrest due to spindle defects). Both support mito function. Hope we can get $$ to learn more. Thanks to Amy and Chloe for their fantastic work.

The very impressive literature on TRiC indicates that there are probably unidentified assembly factors. We show that TRiC assembly subcomplexes first described by the Frydman lab accumulate when MSTO1 is depleted. This is consistent with MSTO1 facilitating assembly of these TRiC subcomplexes!

It’s been fun to learn about TRiC. TRiC is composed of two rings with eight subunits on each ring. When assembled they form a chamber where substrates fold. But how are the rings built? The subunits can’t spontaneously assemble and are structurally very similar, although biochemically distinct.

We reduce TRiC directly and also see mito fragmentation, suggesting that TRiC supports mito function. TRiC is required to fold tubulin and actin, required for mito dynamics, but we see no changes in the levels of these proteins or the cytoskeletal structures when MSTO1 is depleted.

The first thing that happens after MSTO1 depletion is reduced TRiC - an essential cytosolic chaperonin. Days later, the mitochondrial fragmentation phenotype observed in patients is recapitulated. Importantly, TRiC is also reduced in patient cells, suggesting this may contribute to pathophysiology.

We thought that cytosolic MSTO1 would be regulating the proteins that mediate outer membrane fusion (Mfn1 & Mfn2). We build an acute degradation system in vertebrate cells to test this hypothesis. We were wrong!

The science part of science is still fun: www.biorxiv.org/content/10.6... A rare mito disease results from mutations in MSTO1. Patient fibroblasts have short mitochondria due to lower rates of fusion and fewer copies of the mito genome (mtDNA). How is MSTO1 regulating fusion and mtDNA copy #?

My 16yo son loves Greek mythology. We devoured the Percy Jackson series when he was younger. Would you say Silence of the Girls appropriate for us to read together? I’ve seen the Circe title before and was curious but we haven’t tried it yet.

We propose that the energetics, and therefore the proportion in each conformation, can be changed by phosphorylation, binding to other proteins, membrane composition…to either favor or disfavor fusion. Take a look! We’d love to hear what you think.

Using time resolved tmFRET we determined the conformational state of Mfn1 in solution at every step of the catalytic cycle! Biggest surprise: Mfn1 is in equilibrium between two conformations when bound to GDP + Pi.

We know that GTP binding and hydrolysis allosterically regulates how mitofusins on two distinct membranes grab and pull, but exactly how that works remains a mystery. We teamed up with Bill Zagotta and @profsharona.bsky.social to use their amazing transition metal FRET technique to watch Mfn1 move.

It’s out! Huge thanks to our reviewers and the editors at JCB! This collaboration was so fun and Sophie Hurwitz was a science rock star 🌟This story is about the mitofusin paralogs (Mfn1 and Mfn2) - large GTPases in the dynamin superfamily. Their primary role is to mediate outer membrane fusion.
