(11/X) In our view, these results point to most timing variation being cell-intrinsic, with the embryo-level property of developmental “tempo” likely emerging from underlying cell behavior.

(10/X) With a quantitative definition of heterochrony, we can find its molecular drivers. For genes associated with CED across tissues, we see that, rather than being regulated by universally acting pathways, timing shifts depend on multiple processes acting in a cell type-dependent manner.
111
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(9/X) We validated this divergent timing in embryos of both species, showing with live imaging that delayed differentiation of vacuolated cells leads to altered growth dynamics in the medaka notochord. Cell type-specific timing differences explain phenotypic variation!
120
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(8/X) Cross-species CED captured unexpected trends: the notochord shows two peaks, one delayed and one accelerated, corresponding to cell sub-populations in this tissue. The cross-species deviations are beyond timing variation you see within species – something we can cleanly compare with CED.
120
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(7/X) Jess found that some tissues in medaka arrived earlier than expected, some later; not predictable from its overall slower tempo and not easy to pick up with other methods.
120
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(6/X) CED captures the phenomenon of a cell’s “age” being far from the embryo age as a whole. For the within species case (mutants, perturbations), CED pinpoints cells with excess delay or acceleration. In the cross-species case, CED puts a number on S.J. Gould’s modern reframing of “heterochrony.”

Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(5/X) Stage matching using transcriptional + cell count data allows us to represent medaka in “zebrafish time.” We leverage our multi-scale data to ask if, after accounting for differences at the embryo-level, any timing variation remains at the cell-level. We call this “cell-embryo deviation” (CED)
120
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(4/X) Where else is this happening? Is medaka development just slo-mo zebrafish development? These questions were the basis for formally defining a general, quantitative metric for heterochrony across the embryo.
120
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(3/X) Take medaka-specific “stellate cells” with rhythmic contractions – we show this function is built on the foundation of a shared epidermal cell identity and repurposes genes that, in zebrafish, are activated at later stages. S/o @simonknoblich.bsky.social for this incredible characterization.
110
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(2/X) Time-resolved atlases spanning all of development allow us to link timing differences to species-specific phenotypes. The goal is not just “what is this cell?” but “when is this cell?”
110
Mike Dorrity @mwdorr.bsky.social · 19/06/2026
(1/X) Assembling a time- and embryo-resolved sci-RNA-seq atlas of (slow) medaka development from blastula to hatch, we use comparative analysis of (fast) zebrafish to understand global + cell type-specific trends in developmental timing. Excited to get this resource out to the fish community!
