New in PLOS Pathogens 🎉 How does OM-89 (Uro-Vaxom) protect against recurrent UTIs? 🦠 Our paper: it activates lysosomes in bladder epithelial cells, pushing intracellular E. coli into acidic, protease-active compartments and improving antibiotic killing. 🧵 10.1371/journal.ppat.1014448 #UTISky #UPEC

A new study led by @microberamon.bsky.social from 🇬🇧 @oxengsci.bsky.social, with @ucl.ac.uk and @uniofleicester.bsky.social, shows that #phagetherapy could help to reduce recurrent #infections, which happen when the 🦠 bacteria responsible for #UTI s ‘hide’ in the tissue of the #bladder.

3D micro-bladder model to test phage therapy on recurrent UTIs
Research finds that phage therapy can eliminate bacteria hidden deep within bladder wall ‘reservoirs’, which normal antibiotics cannot reach

Ecstatic to have contributed to this wonderful work of my fantastic colleague @microberamon.bsky.social ! Bottom line is that environment, including fluidics, matter for antibiotic treatment which might contribute to treatment failure. Precious collaboration with bioengineers!

(5/6) Most strikingly, flow promoted intracellular bacterial community formation, generating larger intracellular reservoirs within the urothelium.

(4/6) Using P-FLO and our image-analysis pipeline, we observed increased urothelial attachment and enhanced bacterial filamentation under flow conditions.

(3/6) To study infection under more realistic conditions, we developed: plug-flow link organoid (P-FLO). P-FLO incorporates physiological flow into our bladder microtissue model, allowing us to investigate how mechanical forces shape host-pathogen interactions.

(2/6) Quantifying intracellular bacterial communities in 3D models is challenging! We developed a reconstruction and segmentation pipeline that allows us to distinguish intracellular (green) from extracellular (orange) bacteria and quantitatively map bacterial reservoirs within the urothelium 🔬🦠
