Targeting ZMYND8 unleashes IL-2 signalling to override T cell exhaustion - Nature
ZMYND8 suppresses IL-2R–STAT5 signalling by inhibiting p300-mediated transcriptional activation of Il2ra to enforce CD8+ T cell terminal exhaustion, and its deletion boosts effector-like states and markedly enhances antiviral and antitumour immunity.

Exploring synovial fluid immune cell profiles in patient with late-onset ACPA+ and ACPA- RA and PMR showed distinct inflammatory phenotypes among ACPA- individuals but relatively homogeneous inflammatory profiles among ACPA+ individuals ACR Open Rheumatology doi.org/10.1002/acr2...
Synovial fluid immune cell profiles in patients with ACPA-positive late-onset RA, ACPA-negative LORA, and PMR. (A) UMAP plot showing the overall composition of synovial fluid immune cells. (B) Cellular composition of synovial fluid in each patient. (C) Comparison of immune cell proportions among the disease groups. UMAP plot visualizing the landscape of synovial fluid T cell subsets. (E) Relative abundance of T cell subsets in each patient. (F) Comparison of T cell subset frequencies among disease groups. P values were calculated using Kruskal–Wallis test followed by Dunn's multiple comparison test. *P < 0.05, **P < 0.01. Data are presented as mean ± SEM. ACPA, anti–citrullinated protein antibody

The long-lived immune system of centenarians
Nature Reviews Immunology, Published online: 23 April 2026; doi:10.1038/s41577-026-01291-5What is unique about the immune system of people who live to extreme old age? Here the authors describe that centenarians may achieve such longevity through several mechanisms, including limiting the pathological effects of inflammageing and immunosenescence, preservation of immune surveillance, sustained gut microbial diversity and maintenance of intestinal barrier integrity.

COPA syndrome spans multiple organs but is defined by STING in the lung
Nature Reviews Rheumatology, Published online: 07 January 2026; doi:10.1038/s41584-025-01350-6A large European cohort clarifies COPA syndrome as a multi-organ interferonopathy defined by STING-driven interstitial lung disease (ILD). These findings refine diagnosis and treatment and establish COPA syndrome as a model for systemic autoimmune rheumatic disease-associated ILD.

The T cells in these patients showed increased metabolic activity. Elevated levels of inflammatory molecules (IL-6, IL-12, and type I interferon) may be driving the aggressive T cell behavior. #ACR25
