Interplay between systemic inflammation and local platelet–extracellular matrix remodeling in pathological ossification. The model integrates local joint inflammation with systemic activation marked by SAA2, which together establish a microenvironment conducive to ossification. Within this environment, growth factor release and progenitor-cell recruitment are enhanced, and these upstream events converge on increased cell–matrix adhesion and ITGA2B-related intracellular signaling. Systemic inflammatory and platelet–ECM pathways thus make partially overlapping contributions to aberrant bone formation.

A 53-year-old woman presented with fever & right neck pain. She had a history of breast carcinoma and had undergone chemotherapy with sacituzumab govitecan, which required pegfilgrastim. Seven days after the last pegfilgrastim dose, she developed generalized fatigue, fever, and right-sided neck pain
Contrast-enhanced CT did not reveal the origin of the fever on day 10 (A). Follow-up CE-CT on day 14 showed arterial wall thickening, especially in the right common carotid artery (B). Carotid ultrasonography showed thickening of the outer lines around the bifurcation of the right common carotid artery into the external and internal carotid arteries, strongly suggestive of arteritis (C).

Aberrant T cell function contributes to tissue damage and increased susceptibility to infection in SLE. Autoreactive CD4+ T cells differentiate into pathogenic Th1 and Th17 cells, which produce proinflammatory cytokines including IFN-γ and IL-17that directly contribute to tissue inflammation and organ damage. Follicular T helper (Tfh) or peripheral T helper (Tph) cells provide excessive help to B cells through the production of cytokines, including IL-21 and IL-22, and costimulatory signals, promoting autoantibody production. These autoantibodies form immune complexes that deposit in target organs, activate complement, and drive tissue injury.
T cell-targeted therapeutic strategies in SLE. Therapeutic approaches in SLE modulate immune responses through multiple mechanisms that either enhance the suppressive capacity of regulatory T cells, inhibit pathogenic effector T-cell responses, or redirect T cells to eliminate autoreactive B cells. Treg-directed therapies include low-dose IL-2, which preferentially expands and activates Treg cells, and CARTreg cell therapy, which aims to increase Treg cell numbers, stability, antigen specificity, and suppressive function

Initially shown to attenuate glomerulonephritis in a mouse model of ANCA-associated vasculitis in 2014, phase II and III trials quickly followed and avacopan was approved for use across the globe in late 2021 and early 2022. Following release of the Walton report in May 2025, as part of a securities fraud litigation against ChemoCentryx, investigations into the ADVOCATE trial by regulatory bodies began in early 2026.
