And mRNPs are not simply cargo. Super-resolution imaging reveals SON/SRRM2 scaffold networks that organize retained mRNPs and remodel with mRNP load, suggesting that nuclear speckle architecture is tightly coupled to this layer of mRNP quality control.

A set of “retention factors” mediates mRNP retention in nuclear speckles, likely by anchoring stalled mRNPs to the speckle scaffold protein SON.

Nuclear speckles are “hot”, but how do they actually sequester mRNPs? We find that stalled mRNPs are retained in nuclear speckles by a combinatorial RNA-binding protein network. www.biorxiv.org/content/10.6...

A combinatorial pre-mRNP retention factor network couples nuclear speckle architecture to splicing quality control
Cells must prevent incompletely spliced transcripts from entering the cytoplasm, yet how splicing status is recognized and converted into selective nuclear retention remains unclear. Here, we identify...

A bacterial nucleolus for cold adaptation
Compartmentalisation enables cells to spatially organise essential biochemical processes. This is exemplified by the eukaryotic nucleolus, which concentrates the machinery for ribosome biogenesis. Bac...

- and further suggest a possible route in the evolution of cellular organization. Over evolutionary time, transient, stress-induced compartments may have served as intermediates toward constitutive organelles such as the nucleolus.

Our findings point to condensates as a shared organizational principle of ribosome biogenesis across domains of life

The DEAD-box ATPase CsdA, a potent RNA-remodeling enzyme, accumulates and forms condensates at sites of active ribosomal RNA transcription, together with the transcription machinery. And this compartment matters: disrupting it impairs ribosome maturation and cold adaptation.

The heatwave is finally over - just in time for our “Frosty Foci” to finally come online! ❄️🦠 We found that E. coli builds a transient nucleolus-like ribosome biogenesis compartment during cold adaptation, when RNA (mis)folding makes ribosome biogenesis challenging.

Loss of PAIP1 alters cytoplasmic mRNP composition, mRNA stability, and poly(A)-tail homeostasis. In Drosophila embryos, maternal PAIP1 depletion shortens poly(A) tails, delays zygotic genome activation, and causes severe developmental defects.

🚀 New preprint 🥳 Fantastic work by our recent PhD graduate Daan ! How are mRNPs remodeled as they exit the nucleus? We identify PAIP1 as a factor that promotes exchange of the nuclear poly(A)-binding protein PABPN1 for cytoplasmic PABPC1, coupling mRNA export to cytoplasmic mRNP maturation.
