(8/9) We think this integrative model could be relevant to trafficking, plasma-membrane repair, mitochondrial respiration, and other "non-autophagy" phenotypes described by us and others in mammalian cells and other organisms/systems.

(7/9) 💡 We propose that ATG9A and ATG2A/B, while organizing lipid flow for autophagosome and amphisome formation, their loss rewires whole-cell lipid dynamics, potentially driving excess sEV release as a compensatory outlet (= sEVs as markers of autophagy/lysosomal disease?).

(6/9) Whole-cell lipidomics uncovered extensive remodeling in ATG9A- and ATG2A/B-deficient cells: ceramide and neutral-lipid accumulation, phospholipid imbalance, and more LDs which we think leads to the hypersecretion of sEVs because blocking neutral sphingomyelinase normalized EV output ❗

(5/9) Now to some cooler stuff! 🤓 EV proteomics showed depletion of autophagy receptors and ferritinophagy factors (no amphisome = no autophagy cargo in sEVs), with enrichment of RNA-binding proteins and endosomal trafficking regulators.

(4/9) 🔬 Live-cell TIRF microscopy showed more MVE–plasma membrane fusion events, especially in ATG2A/B DKO cells suggesting that the hypersecretory phenotype uses canonical MVE exocytosis machinery ✅

(3/9) The ATG9A/ATG2A/B-deficient cells hypersecreted sEVs 🤔 but failed to load autophagy cargo, revealing a central role for amphisomes, a hybrid organelle born from the fusion between autophagosomes and MVEs.

Carlos Guardia @charlyguardia.bsky.social · 22/07/2026
(2/9) Removing ATG9A or ATG2A/B did not simply block autophagy, it markedly increased secretion of smaller, CD63-enriched sEVs and increased ILVs inside multivesicular endosomes (MVEs). Remarkably, the phenotype was strongest during lysosomal inhibition❕

(1/9) 🚨 Our lab’s first major paper is now out in Nature Communications! We show that ATG9A and ATG2A/B act as lipid-dependent gatekeepers linking autophagosome/amphisome formation to small extracellular vesicle (sEV) secretion.
