Two DNA double helices were directly imaged zipping together for the first time, confirming a more than 20-year-old model. The pairing was precise, groove for groove, but some sequences formed stronger hotspots.

Decades-old DNA mystery solved after strands are captured zipping together for the first time
Researchers have captured the moment two DNA molecules zip together, overcoming their identical negative charges to pair up and revealing a mechanism that has puzzled scientists for more than 20 years.

Supercoiling not only brings DNA to life (and makes it dance) but also changes how key proteins such as #CRISPR interact with it. Thanks @qmsmith.bsky.social for the brilliant collaboration, Sylvia for developing an amazing new #imageanalysis pipeline & @eddierollins.bsky.social for beautiful #AFM.

And finally, we described melting bubbles in highly overtwisted DNA, once again in agreement with biochemical assays from Zechiedrich's lab (6/6)

We also describe the dynamics and diffusion of supercoils. Big end-to-end fluctuations are caused by supercoil reorganization as described by Lipfert and @ecarlon.bsky.social labs (5/6)

We find a new state in negative supercoiling and minimal tension where bubbles form in AT-rich regions irrespective of the underlying DNA topology. They were already detected by Zechiedrich's lab using biochemical assays (bubbles in red) 👇 (3/6)

We provide the conformational phase diagram. We map the regime where bubbles and supercoils are excluded from each other in moderate levels of supercoiling. We name it 'elastic' because it was deduced by rod elastic models (2/6)
